Obviously this is a little break from magic's usual fantasy setting, but just go with it for now. (I tried something similar ages ago at: http://www.magicmultiverse.net/cardsets/57)
We need all sorts of cards: from simple commons that sell superhero flavour to splashy cards that represent iconic heroes.
Do:
Hit as many tropes as possible
Take a broad approach to superhero, include aliens, villains, godzillas, etc if they feel appropriate
See if you can think of any mechanics which might be appropriate for the set!
Don't:
Don't use trademarked superheroes. Have a superhero a bit like superman, but don't call it "superman" and don't try to replicate all the powers exactly.
Get too complicated, don't try to do everything on one card. A big splashy flying hero is fine, it doesn't need indestructible AND direct-damage AND freeze-breath, etc, etc. Make several heros!
I'm going through finals week right now, so I was planning on laying low on Multiverse. But there's no way I'm not contributing to this challenge. If anything this challenge could be considered too broad. When I got done making an equipment, I realized that we could fill this challenge with just superhero equipment and not even scratch the surface of all our options.
Not only Jack's superhero set, but also the Worm cardset, could be taken wholesale as preemptive entries for this challenge. Going for notes not particularly hit by those (that I remember), I created Fire Villain's Lair and Secret Blueprints.
Design a card for a superhero themed set!

Anyhow, I added Ricochet Shield to get us started.
Best of luck with finals!

I've added The Prodigy to my list of Pass the Mantle creatures. Still don't know if it's clear or a good idea, but I like the concept.
