Well, ideally, you'd rename it to be something like "Mosquito Nest" or something. And it could just be a very large mosquito!
I hate mosquitos.
Well, I would think of that one Mosquito as a Swarm. Wouldn't it be otherwise weird that a single mosquito could take out a trained Human Soldier (1/1 token)? ... though rats and squirrels seem pretty proficient at that already.
Also, one could just increase that ability to something like

How does turning it to an enchantment make it less of a flavor fail? I don't get it. You could still activate it only once per turn unless you had flash shenanigans going on.
IMO Mosquito Pool not being a land would be pretty off putting.
I'd like the flavor of this and the original better if it was an enchantment that read:

As a land, it seems odd (though quite understandable, for balance reasons) that my mosquito-infested watering hole only produces one actual mosquito before needing to be refertilized.

Huh, okay. The card I kept thinking was Accumulated Knowledge since the most fascinating facet in this design to me is how it interacts with the following copies of itself.
The main problem is obviously the use of new kind of counter. Unfortunately, I don't see how they could be changed into poison or energy counters without worsening the gameplay experience.
Reminds me of Private Research, and also the verse counter cycle like Rumbling Crescendo.
